Tryptophan for Sleep: Night Waking, Dose and Safety
Sleep & NighttimeArticleA review found a benefit for time awake after falling asleep. Only four articles supplied its pooled analysis, and its dose categories need careful interpretation.

Did the review find that tryptophan helps sleep?
The 2022 Nutrition Reviews1 review collected 18 articles. Data from four entered its random-effects meta-analysis and meta-regression, so 18 articles did not contribute to one pooled result.1
The authors reported less wake after sleep onset, meaning time awake after initially falling asleep. They found no effect on other sleep components. The benefit concerned a specific part of the night.1
This summary follows the published abstract. It does not establish which subgroup of women would benefit or how long an individual should take it.
Does its one-gram category tell me what to take?
The review compared studies using less than 1 gram daily with those using 1 gram or more. The higher-dose category had shorter wake-after-sleep-onset time. That grouping does not identify a universally effective dose, an upper safe limit or a dose suitable for your medicines.1
Before buying a product, write down its L-tryptophan amount per capsule and per suggested serving. Bring the whole ingredient list, including anything else added for sleep. A multi-ingredient capsule cannot be evaluated from the tryptophan amount alone.
Why did an insomnia guideline advise against it?
The American Academy of Sleep Medicine’s2 2017 guideline made a weak recommendation against tryptophan for adults’ sleep-onset or sleep-maintenance insomnia. Its recommendation was based on 250 mg doses. Only one study supplied enough data for its quantitative assessment.2
That study involved 31 people, three interventions and one week of treatment. It compared food-source or pharmaceutical tryptophan, each with carbohydrate, against carbohydrate alone, using sleep diaries. The guideline reported about 9.7 fewer minutes awake after sleep onset, which did not meet its threshold for clinical importance. Harms could not be pooled.2
The guideline predates the later review. The two publications used different evidence sets and dose comparisons. Their findings need those dates and methods attached; neither supplies a personal supplement plan.12
What needs checking before I add a bottle?
NCCIH3 warns that L-tryptophan supplements can cause life-threatening serotonin toxicity when combined with medicines affecting serotonin metabolism. Have a pharmacist review the proposed product alongside all prescriptions, nonprescription medicines and other supplements. A sleep label does not remove that interaction concern.3
For persistent insomnia, NCCIH3 identifies multicomponent cognitive behavioral therapy for insomnia, or CBT-I, as the most strongly recommended treatment. Ask about access to that treatment while reviewing the supplement question.3
Did I Do That in My Sleep? discusses supplement claims in Chapter 10 and advises against adding several sleep products at once. Bring a short record showing when you fall asleep, approximate minutes awake during the night and next-day difficulties. Ask: “Which sleep problem are we treating, and what improvement would justify continuing this approach?”
Educational information, not a supplement dosing recommendation. Ask a pharmacist or clinician about interactions before adding L-tryptophan, particularly if you take medicines affecting serotonin.
